چکیده

A novel type of multicellular spheroids was established and character ized with regard to growth behavior, proliferation, and differentiation. The spheroids were grown from clonal rat rhabdomyosarcoma cells using the spinner flask technique. The cell aggregates showed several unique properties that were different from those observed in most of the spher oids investigated to date. These properties include a non-Gompertzian volume growth; the coexistence of undifferentiated mononuclear cells and of differentiated, myotube-like giant cells with numerous nuclei; a rela tively homogeneous intraspheroidal distribution of proliferating mononuclear cells with thymidine labeling even in the center of spheroids > 1 mm; the absence of necrosis in such large spheroids, and the accumulation of myotube-like cells in the center of these spheroids instead. There was a decrease in the overall proliferative activity of the mononuclear cells with increasing proportion of giant cells in the rhabdomyosarcoma spheroids.

Evaluation of combination effects of 2- methoxyestradiol and methoxyamine on IUdRinduced radiosensitization in glioma spheroids

Background: Glioblastoma is the most common and most malignant cancer of central nervous system. Targeted radiotherapy is an effective method toward its treatment. Iododeoxyuridine (IUdR) is a halogenated thymidine analogue known to be effective as a radiosensitizer in human cancer therapy. In this study we have evaluated the combination effects of 2-Methoxyestradiol, an inhibitor of ...

Fibroblast EXT1-Levels Influence Tumor Cell Proliferation and Migration in Composite Spheroids

BACKGROUND Stromal fibroblasts are important determinants of tumor cell behavior. They act to condition the tumor microenvironment, influence tumor growth, support tumor angiogenesis and affect tumor metastasis. Heparan sulfate proteoglycans, present both on tumor and stromal cells, interact with a large number of ligands including growth factors, their receptors, and structural components of t...

The frequency of PAX3 and PAX7 Mutations in Children with Rhabdomyosarcoma

Abstract Background: Rhabdomyosarcoma is the most common soft tissue sarcoma among children which has two major subtypes: embryonal rhabdomyosarcoma (ERMS) and alveolar rhabdomyosarcoma (ARMS). Distinction between these subtypes is mandatory to choose proper treatment and to determine prognosis. Histopathologic study is the main method, but nowadays molecular studies like PCR are also used...

The inhibition of proliferation and migration of glioma spheroids exposed to temozolomide is less than additive if combined with irradiation.

The aim of this study was to investigate the effect of temozolomide (TZM) in combination with X-rays on proliferation and migration in human glioma spheroids. Multicellular spheroids were derived from GaMg and U87 cell lines. Spheroids were treated with various concentrations of TZM (5 micromol, 0.025 mmol, 0.05 mmol) and irradiation (RT). Proliferation and migration assays were performed. For ...

Rhabdomyosarcoma: Nose Presenting As Second Primary with Carcinoma Tongue

Introduction Presence of two primary malignancies is rare and occurs in 3-5% of the cancer patients. As per our extensive internet research, this is the only reported case of a synchronous sino-nasal embryonal rhabdomyosarcoma with squamous cell carcinoma-tongue. The case report is important because of the rare diagnosis and the challenge we faced in the diagnosis and treatment of the pat...
